Longevity research investigates the twelve hallmarks of aging: genomic instability, telomere attrition, epigenetic drift, loss of proteostasis, deregulated nutrient sensing, mitochondrial dysfunction, cellular senescence, stem cell exhaustion and chronic inflammation. Peptides are used to probe how many of these pathways can be modulated, especially around mitochondrial protection, telomere biology and metabolic tuning.

Mitochondrial protection

In aged cells, cardiolipin in the inner mitochondrial membrane is progressively remodeled, causing electron transport chain leakage and reactive oxygen species buildup. SS-31 class peptides bind cardiolipin to stabilise the inner membrane, restoring ATP capacity and reducing oxidative stress. Studies typically read out maximal respiration, ATP/ADP ratios and ROS levels.

Telomere and pineal axis

Epitalon (Epithalon) is a pineal-derived tetrapeptide (Ala-Glu-Asp-Gly). In cell models it has been correlated with upregulated telomerase activity, and it also appears to entrain melatonin rhythmicity. Research directions include telomere length, cortical rhythmicity and immune markers under long-term low-dose exposure.

Senescence and clearance

Senescent cells keep secreting inflammatory factors (the SASP) and drive tissue aging. Designer peptides such as FOXO4-DRI have shown, in preclinical models, selective induction of apoptosis in senescent cells while sparing healthy ones. This is a central line of senolytic research.
